预处理方式对海带生物质利用性能的影响Effects of Pretreatment Methods on the Utilization Performance of Laminaria japonica Biomass

摘要(Abstract):
【目的】探究预处理方式对海带生物质利用率及产氢量的影响,为实现大型海藻资源化利用提供实验依据。【方法】通过苯酚硫酸法、3,5-二硝基水杨酸法和高效液相色谱法,测定不同预处理的海带水解液中生物质利用率(多糖含量、还原糖含量和单糖的组分及含量)。采集渤海天津海域潮间带污泥作为菌源,不同预处理的海带水解液为混合菌群富集培养基,测定混合菌群的生长和产氢量。【结果】硫酸预处理显著提高了海带生物质利用率(P <0.001),其中多糖质量浓度为(0.71±0.02)mg/L,还原糖质量浓度为(0.34±0.02)mg/mL,主要单糖质量分数为54.01%。混合菌群基于盐酸预处理的海带水解液累积产氢量最高,为(825.68±14.5)mL/g,相比对照组提高3.7倍。干热、湿热和硫酸预处理下,混合菌群利用海带水解液产氢量分别为(222.22±19.8)mL/g、(237.04±23.8)mL/g和(340.74±22.1)mL/g。【结论】以海带水解液为唯一底物时,硫酸预处理方式对提高海带生物质利用率具有显著促进效应;盐酸预处理方式下,富集混合菌群利用海带水解液产氢效率最高,可应用于海带生物质能源化利用。
