罗非鱼干扰素诱导蛋白35基因克隆及表达分析Cloning and Expression Analysis of ifi35 in Nile Tilapia(Oreochromis niloticus)

摘要(Abstract):
【目的】研究尼罗罗非鱼(Oreochromis niloticus)干扰素诱导蛋白35(interferon induced protein 35,ifi35)在免疫反应中的作用。【方法】克隆获得罗非鱼ifi35基因开放阅读框序列(GenBank登录号:XM_003442606;On-ifi35),采用实时荧光定量PCR技术检测On-ifi35在健康罗非鱼不同组织中的分布,以及不同刺激物刺激后的表达。【结果与结论】On-ifi35基因编码区为1125bp,编码374个氨基酸,理论分子质量为42.79ku,等电点为5.01。On-ifi35含有1个LZD结构域和2个Nmi/IFP 35 domain (NID)。多序列比对发现On-ifi35与其它物种的ifi35氨基酸序列同源性介于35.23%~93.32%之间,且与斑马拟丽鱼同源性最高。系统进化分析发现,On-ifi35与斑马拟丽鱼的ifi35聚为一支。实时荧光定量PCR分析显示,On-ifi35在健康罗非鱼各组织中均有表达,其中在脾脏组织中表达量最高,其次是胸腺、肝脏、心脏、肌肉、肠道、体肾、脑、鳃、头肾,在皮肤中表达量最低。经无乳链球菌和PolyI:C刺激后,On-ifi35表达量在脾脏、肠道、体肾和头肾中均发生显著性变化且呈现出显著的时序性。
