亨氏马尾藻岩藻聚糖结构鉴定及其对氧化型低密度脂蛋白诱导HMVEC损伤的保护作用Structural Identification of Sargassum henslowianum C. Agardh Fucoidan and Its Protective Effect on HMVEC Damage Induced by Oxidized Low Density Lipoprotein

摘要(Abstract):
【目的】探讨4种亨氏马尾藻岩藻聚糖的结构及其对氧化型低密度脂蛋白诱导HMVEC损伤的保护作用。【方法】以亨氏马尾藻岩藻聚糖(FD1)为原料,过Sepharose Cl-6B琼脂糖凝胶柱层析得到组分FS11,对FS11进行化学组成分析和单糖分析,测定F、FD1、FS1、FS11等4种岩藻聚糖的相对分子质量、糖链构象和红外光谱,进行结构鉴定;采用MTT法测定3种岩藻聚糖对HMVEC存活率的影响,研究4种岩藻聚糖对氧化型低密度脂蛋白诱导HMVEC损伤的保护作用。【结果】与F、FD1、FS1比,FS11硫酸基和多糖含量最高。FS11中岩藻糖含量高达52.744%;F、FD1、FS1、FS11的相对分子质量分布分别为474 576、459 032、201 285、190 049;I-KI紫外扫描和刚果红实验表明岩藻聚糖具有复杂树状分支和多股螺旋结构;红外光谱结果表明各组分均有O—H、C—H多糖特征官能团、S==O活性特征官能团;加药培养24 h,质量浓度为500μg/mL和1 000μg/mL的4种岩藻聚糖(F、FD1、FS1、FS11)均能显著减弱受氧化型低密度脂蛋白损伤的HMVEC的增殖抑制(P<0.01)。【结论】尾藻岩藻聚糖对氧化型低密度脂蛋白诱导的HMVEC损伤具有保护作用。
