半叶和亨氏马尾藻岩藻聚糖理化性质及其抗氧化抗肿瘤活性Physicochemical Properties and Antioxidant and Antitumor Activities of Fucoidan from Sargassum hemiphyllum and Sargassum henslowianum

摘要(Abstract):
【目的】比较半叶马尾藻岩藻聚糖(Fb)和亨氏马尾藻岩藻聚糖(Fh)的理化性质及其抗氧化和抗肿瘤活性,探讨马尾藻岩藻聚糖的理化性质与其抗氧化和抗肿瘤活性的构效关系。【方法】运用超声热水浸提法提取Fb和Fh,进行化学组成和紫外分析,通过红外光谱进行结构鉴定;通过测定其清除DPPH自由基的能力和总抗氧化能力(FRAP法)比较Fb和Fh的体外抗氧化活性;运用MTT法测定其抗肿瘤活性。【结果】Fb中总糖(75.33±3.99)%、L-岩藻糖(15.63±3.37)%和硫酸基(29.16±0.86)%的含量均高于Fh。紫外扫描表明Fb和Fh不含核酸,仍含少量蛋白;红外光谱结果表明Fb和Fh均有O—H、C—H多糖特征官能团、S=O活性特征官能团;体外抗氧化结果表明,Fb的DPPH自由基清除能力和总抗氧化能力均强于Fh,并呈剂量依赖性,表明Fb抗氧化活性强于Fh。MTT结果表明,随着Fh和Fb质量浓度增大,HepG2细胞存活率呈下降趋势,且当Fb和Fh质量浓度为8 mg/mL时,HepG2细胞的存活率分别为(21.17±2.54)%、(27.21±3.69)%,说明Fb的抗肿瘤活性也强于Fh。【结论】马尾藻岩藻聚糖的抗氧化能力和抗肿瘤活性与其L-岩藻糖、硫酸根的含量有关,且抗肿瘤活性与其抗氧化能力也相关。
