洗脱剂极性对沉积物长链烯酮硅胶柱层析效果的影响Influence of Eluent Polarity on Separation of Sediment Long-Chain Alkenones in Silica Gel Column Chromatography

摘要(Abstract):
目的】研究洗脱溶剂极性对海洋沉积物中C_(37)和C_(38)烯酮的柱层析分离效果及对重建古海表温度的影响,探寻能完全洗脱长链烯酮而不引入过多杂质的洗脱溶剂配比。【方法】通过改变正己烷(H)与二氯甲烷(D)体积比来调节洗脱液极性,按极性从小到大对标准样品进行洗脱,并计算每种烯酮分子的洗脱量。【结果】当V_H∶V_D=8∶1时烯酮开始被洗脱出,到1∶2时可被完全洗脱。烯酮C_(38:2)Et最先被洗脱出,而C_(37:3)最后被洗脱出。各烯酮分子被洗脱量的峰值出现在不同极性的洗脱液,按极性排序为C_(38:2)Et
