马氏珠母贝细胞周期分裂基因CDC45基因特征、表达量与性状相关分析Molecular Characterization of Cell Division Cycle Gene 45 and Its Relationship Between Its Relative Expression and Growth Traits in Pinctada fuctada martensii

摘要(Abstract):
采用RACE技术克隆了马氏珠母贝细胞周期分裂基因45(Pm-CDC45)全长序列,利用荧光定量PCR检测该基因在闭壳肌、鳃、性腺与外套膜组织的表达量,并估计基因表达量与金黄壳色第3代选育群体壳高性状的相关性。结果表明:Pm-CDC45基因序列全长为2 091 bp,5′非编码区(5′UTR)为157 bp,3′非编码区(3′UTR)为34 bp,其中开放阅读框为1 967 bp,编码655个氨基酸;Pm-CDC45在各组织的相对表达量存在显著差异(P<0.05),其中闭壳肌表达量最高,鳃和性腺为其次,外套膜最低;Pm-CDC45基因相对表达量与选育群体壳高性状与存在显著的正相关(r=0.531,P<0.05);马氏珠母贝Pm-CDC45为影响壳高性状基因。
